Marie-José Goumans is hoogleraar cardiovasculaire celbiologie bij de afdeling cel en chemische biologie. In 2009 trad Marie-José Goumans toe tot de Jonge Akademie. In 2004 ontving zij een VIDI beurs. De focus van het onderzoek is gericht op het gebruik van cardiale (stam)cellen en signaaltransductie-paden die noodzakelijke zijn voor cardiovasculair herstel. Het ontrafelen van deze processen is belangrijk voor zowel “tissue engineering” van hart en bloedvaten in een schaaltje, (stam)cel transplantatie, als ook voor herstel van binnenuit door paracriene signalen. Het ontwikkelen van het hart en de bloedvaten is een nauwlettend proces. Cellen delen zich om er meer van te krijgen, en reageren op groeifactor signalen uit de omgeving. Interactie tussen cellen en hun omgeving is zeer bepalend voor overleving en differentiatie. Zowel het hart als ook de bloedvaten zijn gebaat bij een optimale regulatie van deze groeifactor, niet alleen tijdens de embryonale ontwikkeling, maar ook bij volwassenen. Als de balans scheef is, zowel teveel, als ook te weinig signalering, zorgt dat voor veel problemen. Groeifactoren die belangrijk zijn voor de differentiatie van stamcellen naar hart en bloedvatcellen in een schaaltje als ook de vorming van hart en bloedvaten in het zich ontwikkelende embryo behoren tot de transformerende groeifactor beta (TGF-β) familie. Mutatie in het TGF-β signaaltransductie pad liggen ten grondslag aan de erfelijke cardiovasculaire ziektes pulmonale arteriële hypertensie en de ziekte van Rendu Osler Weber. Verder draagt een verstoorde TGF-β signalering bij aan een toename van fibrose in het hart en de ontwikkeling van hartfalen.
Voor het bestuderen van herstel van hartweefsel wordt onderzoek gedaan naar (stam)cellen die zich in het hart bevinden. Uit humane hartbiopten worden cardiale progenitor cellen CPC geïsoleerd. Deze hartspiervoorlopercellen zijn in staat om, na transplantatie in hart na een infarct, de functie te verbeteren. Naast de CPC is ook de epicardiale cel (EPDC) belangrijk voor een goed herstel van het hart na schade. De EPCD is zelf niet in staat om nieuwe hartspiercellen te vormen maar geeft steun en instructie aan de herstellende hartwand. Deze twee humane cardiale cel populaties bieden de mogelijkheid ‘hartweefseltjes’ te genereren om te kijken hoe cellen zich in 3D gedragen. Groeifactoren en exosomen die worden uitgescheiden door de cardiale cellen hebben een positief effect of weefsel herstel. CPCs en EPDCs zijn een uitstekend cel model om de paracrine signalen die belangrijk zijn voor hartreparatie, zoals TGF-β, te bestuderen. Een thema is dan ook of via endogene activatie van cellen of in vitro tissue engineering, de regeneratieve capaciteit van het hart kan worden bevorderd.
Wetenschappelijke carrière
Marie-José Goumans, heeft haar tijdens haar promotieonderzoek gedaan op het Hubrecht Laboratorium in Utrecht, Nederland, naar de rol van TGF-beta tijdens de ontwikkeling van hart en bloedvaten. Ze heeft haar promotie in 1999 afgerond met het verdedigen van haar proefschrift getiteld: Functional analysis of TGF-β signalling in early mouse development. Na haar promotie heeft ze als post-doc bij het Ludwig Institute for Cancer research in Uppsala, Zweden en het Nederlands Kanker Instituut, onderzoek gedaan om te begrijpen TGF-β het gedrag van bloedvaten beïnvloed. In 2003 begon ze haar eigen groep bij de afdeling cardiologie in het UMC Utrecht met als centraal thema de regeneratieve capaciteit van het hart en de rol van cardiale voorloper cellen, waarvoor ze in 2004 een VIDI-beurs ontving. In 2008 verhuisde Marie - José Goumans met haar groep naar de afdeling Moleculaire Cel Biologie in het LUMC waar ze haar onderzoek naar stamcellen en herstel van hart en bloedvaten voortzetten, met als focus de invloed van verstoorde TGFβ signalering. In 2009, werd ze lid van de jonge akademie (DJA) van de KNAW. In Februari 2012 werd Marie-José benoemd tot hoogleraar moleculaire cardiovasculaire celbiologie aan de universiteit Leiden. Op 2 November 2012 hield zij haar inaugurele reden met als titel: “Hart voor de wetenschap”.
